Description
What it is and why it is different
Retatrutide is a triple agonist: it simultaneously activates three receptors: GLP-1, GIP and glucagon. It is the first compound in its class to combine three metabolic pathways in a single molecule, with a synergistic effect that research suggests is greater than that of single or dual agonists.
While semaglutide activates only the GLP-1 receptor and tirzepatide activates GLP-1 + GIP, retatrutide also adds the glucagon receptor: this means it not only modulates appetite and blood glucose, but also energy expenditure. Glucagon stimulates lipolysis and thermogenesis: it is the receptor that tells the body to burn energy, not just store it.
Why the glucagon receptor matters
Glucagon is the hormone antagonist of insulin. While insulin stores energy, glucagon mobilizes it. Activating the glucagon receptor means:
- Accelerated lipolysis: adipose tissue releases fatty acids for energy use
- Thermogenesis: the body produces more heat, a signal of energy expenditure
- Redistribution of energy substrate: less accumulation, more utilization
Retatrutide combines this effect with appetite regulation (GLP-1) and improved insulin sensitivity (GIP). Three mechanisms, one molecule.
